Numerous reports have demonstrated that CD4(+)CD25(+) regulatory T cells (Tregs) from individuals with a range of human autoimmune diseases, including type 1 diabetes, are deficient in their ability to control autologous proinflammatory responses when compared with nondiseased, control individuals. Treg dysfunction could be a primary, causal event or may result from perturbations in the immune system during disease development. Polymorphisms in genes associated with Treg function, such as IL2RA, confer a higher risk of autoimmune disease. Although this suggests a primary role for defective Tregs in autoimmunity, a link between IL2RA gene polymorphisms and Treg function has not been examined. We addressed this by examining the impact of an IL2RA haplotype associated with type 1 diabetes on Treg fitness and suppressive function. Studies were conducted using healthy human subjects to avoid any confounding effects of disease. We demonstrated that the presence of an autoimmune disease-associated IL2RA haplotype correlates with diminished IL-2 responsiveness in Ag-experienced CD4(+) T cells, as measured by phosphorylation of STAT5a, and is associated with lower levels of FOXP3 expression by Tregs and a reduction in their ability to suppress proliferation of autologous effector T cells. These data offer a rationale that contributes to the molecular and cellular mechanisms through which polymorphisms in the IL-2RA gene affect immune regulation, and consequently upon susceptibility to autoimmune and inflammatory diseases.  相似文献

Enterohaemorrhagic Escherichia coli O157 : H7 is a bacterial pathogen that can cause haemorrhagic colitis and haemolytic uremic syndrome. In the primary reservoir host, cattle, the terminal rectum is the principal site of E. coli O157 colonization. In this study, bovine terminal rectal primary epithelial cells were used to examine the role of H7 flagella in epithelial adherence. Binding of a fliCH7 mutant O157 strain to rectal epithelium was significantly reduced as was binding of the flagellated wild-type strain following incubation with H7-specific antibodies. Complementation of fliCH7 mutant O157 strain with fliCH7 restored the adherence to wild-type levels; however, complementation with fliCH6 did not restore it. High-resolution ultrastructural and imunofluorescence studies demonstrated the presence of abundant flagella forming physical contact points with the rectal epithelium. Binding to terminal rectal epithelium was specific to H7 by comparison with other flagellin types tested. In-cell Western assays confirmed temporal expression of flagella during O157 interaction with epithelium, early expression was suppressed during the later stages of microcolony and attaching and effacing lesion formation. H7 flagella are expressed in vivo by individual bacteria in contact with rectal mucosa. Our data demonstrate that the H7 flagellum acts as an adhesin to bovine intestinal epithelium and its involvement in this crucial initiating step for colonization indicates that H7 flagella could be an important target in intervention strategies.  相似文献

The cueO gene of Escherichia coli encodes a multi-copper oxidase, which contributes to copper tolerance in this bacterium. It was observed that a cueO mutant was highly sensitive to killing by copper ions when cells were grown on defined minimal media. Copper sensitivity was correlated with accumulation of copper in the mutant strain. Growth of the cueO mutant in the presence of copper could be restored by addition of divalent zinc and manganese ions or ferrous iron but not by other first row transition metal ions or magnesium ions. Copper toxicity towards a cueO mutant could also be suppressed by addition of the superoxide quencher 1,2-dihydroxybenzene-3,5-disulfonic acid (tiron), suggesting that a primary cause of copper toxicity is the copper-catalyzed production of superoxide anions in the cytoplasm.  相似文献

Endocytosed antigens are proteolytically processed and small amounts of peptides captured by class II MHC molecules. The details of antigen proteolysis, peptide capture and how destruction of T-cell epitopes is avoided are incompletely understood. Using the tetanus toxin antigen, we show that the introduction of 3-6 cleavage sites is sufficient to trigger a partially unfolded conformation able to bind to class II MHC molecules. The known locations of T-cell epitopes and protease cleavage sites predict that large domains of processed antigen (8-35 kDa) are captured under these conditions. Remarkably, when antigen is bound to the B-cell antigen receptor (BCR), processing can trigger a concerted 'hand-over' reaction whereby BCR-associated processed antigen is captured by neighbouring class II MHC molecules. Early capture of minimally processed antigen and confinement of the processing and class II MHC loading reaction to the membrane plane may improve the likelihood of T-cell epitope survival in the class II MHC pathway and may help explain the reciprocal relationships observed between B- and T-cell epitopes in many protein antigens and autoantigens.  相似文献

Trans‐equatorial avian migrants tend to breed, moult and migrate – the main energy‐requiring events in their lifecycle – at different times. Little is known about the relationship between wing moult and pre‐migratory fuelling in waders on their non‐breeding grounds, where time is less constrained than during their brief high‐latitude breeding season. We determined age‐related strategies of Wood Sandpipers Tringa glareola to balance the energetic demands of primary moult against pre‐migratory fuelling in southern Africa by analysing body mass and primary moult at first capture of 1721 birds mist‐netted in 1972–96 at waterbodies in Zimbabwe. Adults moulted all their primaries in August–December, but immatures underwent a supplemental moult of varying numbers of outer primaries in December–April, close to departure. We used locally weighted linear regression to estimate trends in Wood Sandpiper body mass from 1 July to 1 May. They maintained low mass from arrival in July–September to February–early March. Adults fuelled from 10 February to 1 May at a mean rate of 0.25 g/day (sd = 0.16). Most adults (98%) began fuelling 10–75 days after completing primary moult. Immatures fuelled from 4 March to 13 April at 0.24 g/day (sd = 0.14). They used varying strategies depending on their condition: a brief gap between moult and fuelling; an overlap of these processes near departure, leading to slower fuelling; or skipping fuelling altogether and staying in southern Africa for a ‘gap year’. Immatures moulting three or five outer primaries fuelled more slowly than post‐moult birds. Immatures moulting four outer primaries started fuelling 3 weeks later but at a higher rate than did post‐moult birds of this group. In post‐moult immatures, the later they ended moult, the later and faster they fuelled. The heaviest adults and immatures using all moult patterns accumulated fuel loads of c. 50% of lean body mass, and could potentially cross 2397–4490 km to reach the Great Rift Valley in one non‐stop flight. Immatures were more flexible in the timing and extent of moult and in the timing and rate of fuelling than adults. This flexibility enables inexperienced Wood Sandpipers to cope with inter‐annual differences in feeding conditions at Africa's ephemeral inland waterbodies.  相似文献

During 2002–2007, we assessed the status of the Purple Heron Ardea purpurea in Numidia, Algeria by surveying all the major wetlands in the region. We located six distinct breeding sites; four of these were not previously known. We also investigated nest site selection and determined the species’ reproductive success at Dakhla, a dunary pond during two successive years (2006 and 2007). The egg laying period was comparable to that reported for southern Europe (March-May) and a seasonal change of breeding success was recorded. A growth curve was derived for developing nestlings and the impact of an ectoparasite (mite) infestation upon nestling's growth was considered. We also examined 73 food boluses regurgitated by nestlings and identified 329 prey items. Fish, mainly Gambusia holbrookii and Cyprinus carpio, dominated by mass, whereas insects, mainly aquatic Coleopteran larvae and aeshnids (Odonata), were the most frequent prey (67.1%). Loss of habitat is identified as a major threat to the future of colonial herons in Algeria.  相似文献
